Yes. At minimum, you boot the system with a disk in, and it will treat it as if it were a fixed hard drive.

I don't know if any version of Iomega's ZIP Tools or "IomegaWare" will work with the Jaz drive under System 6, though. (I know there's one version of ZIP Tools that supports System 6, but I don't know if it supports a Jaz drive.

Does the disk have to be formated first?

It would help, since System 6's HD SC Setup won't be able to partition it. But it *MIGHT* pop up as "unformatted disk, would you like to format?"
